The overall effect of miRNAs on L1 starvation survival is expected to be significantly stronger than that reflected by the data in Fig. These results suggest that miRNAs act in the intestine, and possibly in other tissues, to promote L1 starvation survival. However, the mechanisms that coordinate the long-term survival, overall developmental arrest, and reinitiation remain to be investigated.
The transcript level of unc-31 was increased in mir-71(lf) worms, compared with that of wild-type controls that were normalized to the value of 1. MiR-71 represses the expression of age-1 and unc-31 through the actions on their 3′UTR, but miR-71 is not required for arresting M cell division during L1 diapause. We found that the poor survival rate of daf-16(mu86)(lf) was further decreased by mir-71(lf) (Fig. 2C), consistent with the notion that a portion of miR-71 activities regulate genes that act in parallel to UNC-31–mediated InsR/PI3K signaling for long-term survival during L1 diapause. Despite our time away from alcohol, there are moments or even extended periods of time when we feel like a life with alcohol isn’t so bad, that we can moderate. Furthermore, worms that are long-lived due to dietary restriction or decreased mitochondrial respiratory rates are short-lived during L1 diapause, suggesting that the mechanisms controlling L1 starvation survival are different at least in some aspects from those controlling aging (3). We provide evidence that miRNA miR-71 is not required for the animals’ entry into L1 diapause, but plays a critical role in long-term survival by repressing the expression of insulin receptor/PI3K pathway genes and genes acting downstream or in parallel to the pathway. We need to be promoting better-quality games that do more to help people have fun, and we need to be modelling and making room for healthy play that promotes pro-social behaviour. That means running programs, preferably having staff who can teach the games, having services that help people find others who can teach and/or are interested in playing, and establishing norms for play in the library space in the hopes of (a) offering that safe, and potentially healing, interaction; (b) having those norms spread.
However, we found that the reporter transgene with the lin-42 3′UTR was significantly repressed in wild-type worms, but derepressed in the mir-71(lf) worms (Fig. 4 H and I). In starved L1 worms, we detected only a slight increase in the mRNA level of hbl-1 in mir-71 mutants compared with that in wild type (∼10%), which may not be biologically significant. These results indicate that miR-71 plays a significant role in larval development of animals recovering from L1 diapause and likely does so by regulating the expression of components of the insulin receptor/DAF-16 pathway, as well as factors acting downstream, or in parallel to, DAF-16. If this were true, the starved mir-71(lf); daf-16(lf) double-mutant worms should show a slow growth phenotype similar to that of daf-16(lf) worms, but no specific VPC timing defect.

However, when newly hatched L1 worms encounter an environment with no food, developmental programs arrest and the worm enters L1 diapause. The presented results indicate that interactions between multiple miRNAs and likely a large number of their mRNA targets in multiple pathways regulate the response to starvation-induced L1 diapause. And we need to take seriously that, just as people’s literary diet matters, so does their ludic diet. In fact I still have a ton to say, about play anxiety and devaluation of play, which I’ll have to put into separate posts over the next few days.
Furthermore, miR-71 plays a prominent role in developmental recovery from L1 diapause partly through repressing the expression of certain heterochronic genes. Here we show that compromising overall microRNA (miRNA) functions or mutating certain individual miRNAs impairs the long-term survival of nematodes during starvation-induced L1 diapause. But offering people opportunities to explore games and play in a safe, non-abusive, self-directed context like a library seems to me to matter, and for folks who are more likely than most to have social anxieties, finding those opportunities can be extremely hard. This certainly accords with the rise of therapists using games as therapy – a web search for “Dungeons & Dragons therapy” shows the recent explosion in ways in which people are finding healing in play. That means that when sales are up, more people are playing this and other older, less-fun games instead of games with the advantage of more than a century’s advancements in the art and science of play, including co-operative games, social games, nomic games, and more.
